    Efficiency of organic pesticides, turmeric (Curcuma longa) and neem (Azadirachta indica) against dry fish beetle (Dermestes sp.) during storage condition

    Efficacy of turmeric (Curcuma longa) and neem (Azadirachta indica) or repelling dry fish insects (Dermestes sp. adult and larva) was investigated on the basis of their repellency rate, storage quality, sensory attributes of turmeric and neem treated dry fish and their cooked products. Pesticides prepared from turmeric and neem, were used in the form of extract and powder. The extracts were tested at the concentrations of 2%, 3%, 5% for turmeric and 3%, 5%, 7% for neem. Turmeric and neem powder were used for treatment at the rate of 2.0%, 3%, 4.0% upon 100 g dry fish. Repellency rate of Dermestes sp., adults was highest, where larvae showed mere resistance against herbal pesticides. Pesticides prepared from turmeric were more repellent than neem pesticides. Control and treated dry fish had almost similar sensory attributes after the 2nd month. However, odor, texture, general appearance and insect occurrence of control dry fish decreased significantly after 4th month of storage but all those parameter of treated dry fish were nearly unchanged. However, sensory quality of dry fish treated with turmeric and neem were remain unchanged after the 4th month. The results suggested that 3% extract and powder of turmeric or neem at the rate of 5% extract and 3% powder can be used as suitable field doses for direct application in dry fish to prevent insect infestation during storage of dry fish products

    Influence of salt and herbal substance on the drying and reconstitution performance of Bombay duck, Harpodon nehereus

    Effects of salt, chili and turmeric powder on the production of high quality dried fish products from Bombay duck (Harpodon nehereus) under open sun drying was investigated. Five different types of dried products (T1-T5) were produced and studied. Moisture content in T3 decreased more rapidly to below 16% within 28 hrs of drying compared to those treated with other treatments (temperature varies from 24.6°C to 34°C and relative humidity varies between  60% and 48%) . Drying process was very slow in control samples where it took 32 hrs for reaching the moisture level to 18.75%. Both turmeric and chili powders had strong repellency effect against insect infestation. Bombay duck treated with salt and herbal products were less infested by the blowfly whereas samples dried under control treatment were severely infested by blowfly

    Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field

    Testing a global standard for quantifying species recovery and assessing conservation impact.

    Recognizing the imperative to evaluate species recovery and conservation impact, in 2012 the International Union for Conservation of Nature (IUCN) called for development of a "Green List of Species" (now the IUCN Green Status of Species). A draft Green Status framework for assessing species' progress toward recovery, published in 2018, proposed 2 separate but interlinked components: a standardized method (i.e., measurement against benchmarks of species' viability, functionality, and preimpact distribution) to determine current species recovery status (herein species recovery score) and application of that method to estimate past and potential future impacts of conservation based on 4 metrics (conservation legacy, conservation dependence, conservation gain, and recovery potential). We tested the framework with 181 species representing diverse taxa, life histories, biomes, and IUCN Red List categories (extinction risk). Based on the observed distribution of species' recovery scores, we propose the following species recovery categories: fully recovered, slightly depleted, moderately depleted, largely depleted, critically depleted, extinct in the wild, and indeterminate. Fifty-nine percent of tested species were considered largely or critically depleted. Although there was a negative relationship between extinction risk and species recovery score, variation was considerable. Some species in lower risk categories were assessed as farther from recovery than those at higher risk. This emphasizes that species recovery is conceptually different from extinction risk and reinforces the utility of the IUCN Green Status of Species to more fully understand species conservation status. Although extinction risk did not predict conservation legacy, conservation dependence, or conservation gain, it was positively correlated with recovery potential. Only 1.7% of tested species were categorized as zero across all 4 of these conservation impact metrics, indicating that conservation has, or will, play a role in improving or maintaining species status for the vast majority of these species. Based on our results, we devised an updated assessment framework that introduces the option of using a dynamic baseline to assess future impacts of conservation over the short term to avoid misleading results which were generated in a small number of cases, and redefines short term as 10 years to better align with conservation planning. These changes are reflected in the IUCN Green Status of Species Standard

    Japanese Spanish mackerel (JSM) (Scomberomorus niphonius) is a marine fish species containing health-beneficial polyunsaturated fatty acids (PUFAs). In the present study, the quality of JSM by-products oils extracted by supercritical CO2 (SC-CO2) and organic solvent extraction was compared in terms of physico-chemical properties of the oils. Eicosapentaenoic acid (EPA) is one of the important polyunsaturated fatty acids present in SC-CO2-extracted skin and muscle oil 5.81 ± 0.69% and 4.93 ± 0.06%, respectively. The amount of docosahexaenoic acid (DHA) in SC-CO2-extracted skin and muscle oil was 12.56 ± 0.38% and 15.01 ± 0.28%, respectively. EPA and DHA are considered as important PUFAs for the development of brain function and the prevention of coronary heart diseases. Extracted oils showed considerable antioxidant activity. In the obtained oils, atherogenic index (AI) values varied from 0.72 to 0.93 and thrombogenic index (TI) ranged from 0.75 to 0.92, which is considered an acceptable level. Fatty acid composition, bio potentiality, thermogravimetric, and vitamin D analysis showed that oils extracted from JSM by-products can be a good source of oil for application in food, pharmaceutical and cosmetic industries. Therefore, the present research revealed the potentiality of green valorisation of S. niphonius by-products as a possible sustainable approach for targeting the era of zero waste

    Small Indigenous Fish: A Potential Source of Valuable Nutrients in the Context of Bangladesh

    Peoples can readily and affordably obtain small indigenous fish species (SIS), with a maximum length of 25 cm, that can be found in niches in small bodies of water (such as canals, ponds, wetlands, streams, etc.). SIS contribute valuable and significant macro- and micronutrients in Bangladesh. SIS are excellent suppliers of vitamins and minerals. The main micronutrients are copper, iodine, iron, selenium, chromium, and zinc, which can help prevent mineral deficiency and enzymatic response in the human body. SIS, such as the mola (Amblypharyngodon mola), cotio (Osteobrama cotio), darkina (Esomus danricus), etc., are easily digestible by the human gut. Nutrients of these fish are quickly accessible by the intestine. These fish are comparatively cheap and easy to capture and promote the presence of particular vitamins in SIS, including A, D, E, and K, as well as vitamin B components. This review paper focuses on the SIS nutritional contribution and their efficacy of preventing malnutrition in developing countries such as Bangladesh and the whole world

    Immunomodulatory Potency of Eclipta alba (Bhringaraj) Leaf Extract in Heteropneustes fossilis against Oomycete Pathogen, Aphanomyces invadans

    Aphanomyces invadans is an aquatic oomycete pathogen and the causative agent of epizootic ulcerative syndrome (EUS) in fresh and brackish water fish, which is responsible for severe mortalities and economic losses in aquaculture. Therefore, there is an urgent need to develop anti-infective strategies to control EUS. An Oomycetes, a fungus-like eukaryotic microorganism, and a susceptible species, i.e., Heteropneustes fossilis, are used to establish whether an Eclipta alba leaf extract is effective against the EUS-causing A. invadans. We found that treatment with methanolic leaf extract, at concentrations between 50&ndash;100 ppm (T4&ndash;T6), protects the H. fossilis fingerlings against A. invadans infection. These optimum concentrations induced anti-stress and antioxidative response in fish, marked by a significant decrease in cortisol and elevated levels of superoxide dismutase (SOD) and catalase (CAT) levels in treated animals, as compared with the controls. We further demonstrated that the A. invadans-protective effect of methanolic leaf extract was caused by its immunomodulatory effect and is linked to the enhanced survival of fingerlings. The analysis of non-specific and specific immune factors confirms that methanolic leaf extract-induced HSP70, HSP90 and IgM levels mediate the survival of H. fossilis fingerlings against A. invadans infection. Taken together, our study provides evidence that the generation of anti-stress and antioxidative responses, as well as humoral immunity, may play a role in protecting H. fossilis fingerlings against A. invadans infection. It is probable that E. alba methanolic leaf extract treatment might become part of a holistic strategy to control EUS in fish species
